Anti-melanogenic Effect of Gamma-irradiated Sericin via p38/MITF Pathway in Melan-a Cell

Abstract
Silk sericin is a water-soluble protein, which derived from silkworm Bombyx mori. Recently, several studies have recognized the various biological activities of sericin. However, studies on the structural changes and physiological activities of silk sericin using the gamma irradiation had not been well addressed. In this study, we evaluated the anti-melanogenic effects of gamma-irradiated sericin by using Melan-a cells. First, both sericin without gamma irradiation and gamma-irradiated sericin treatment at 100 ¥ìg/mL significantly decreased melanin content more than 20%, compared to negative control and especially, 25 kGy gamma-irradiated sericin (GIS) decreased melanin content by 40%, compared to negative control. And 25 kGy GIS significantly decreased microphthalmia- associated transcription factor (MITF) and tyrosinase related protein (TRP)-2 protein expression, compared to negative control, which was similar to the expression of arbutin. Also, in the mitogen-activated protein kinase (MAPK) pathway, which is upstream pathway of MITF, 25 kGy GIS decreased the expression of phosphorylated p38. Taken together, GIS showed anti-melanogenic effect in Melan-a cells by inhibition of p38-dependent MITF pathway.
